bright-field microscopy of fixed tissue stained using carmine or hematoxylin

nucleus appears as a restricted compartment with clustered or completely red (carmine) or black (hematoxylin) pigmentation

transmission electron microscopy, stained with heavy metals

nucleus appears as a membrane-bound compartment scattered irregularly with electron-dense clusters of varying size to complete electron-dense staining
